Poor sleep is one of the most common complaints clinicians see, and one of the most supplemented. The trouble is that the supplement aisle doesn’t distinguish between compounds with strong clinical data and those riding on mechanism alone — this protocol does.
The six supplements here address three distinct problems: trouble falling asleep, poor sleep quality and depth, and stress-driven wakefulness. Not everyone needs all six — match the supplement to the problem you’re actually dealing with rather than stacking all of them and hoping. Dose and form matter enormously: magnesium oxide won’t do what glycinate does, and 10 mg of melatonin won’t outperform 0.5 mg (it will likely work worse). Each entry specifies what matters.
A note on scope: This protocol is for otherwise healthy adults with general sleep complaints. It is not designed for shift workers, people with diagnosed sleep disorders (such as obstructive sleep apnea or restless leg syndrome), or those on medications with sedative effects, without first speaking to a clinician. If you’ve been struggling with sleep for more than three months, discuss a sleep study with your doctor before adding supplements.

Glycine Moderate
Glycine is a non-essential amino acid with a well-characterized mechanism: it lowers core body temperature by increasing peripheral blood flow — a key trigger for sleep onset — and acts on NMDA receptors in the suprachiasmatic nucleus, the master circadian clock. Part 3: Stress-Driven Sleep Disruption
Ashwagandha (KSM-66) Moderate

Apigenin Low

Most supplements in this protocol are well-tolerated and have limited interaction profiles at standard doses. The two to watch carefully are melatonin and ashwagandha.
- Melatonin + sedatives or benzodiazepines: Additive central nervous system (CNS) depression. Avoid combining without clinician guidance; if used together, lower the melatonin dose.
- Melatonin + warfarin (and other anticoagulants): Melatonin may enhance anticoagulant effects. Monitor INR more closely if added to an anticoagulant regimen.
- Melatonin + fluvoxamine: Fluvoxamine significantly increases melatonin blood levels via CYP1A2 inhibition. Use the lowest effective melatonin dose if co-prescribed.
- Ashwagandha + thyroid medications (levothyroxine): Ashwagandha may increase thyroid hormone levels. Thyroid function should be monitored if used chronically alongside thyroid medication.
